Patralekhaa starred in the streaming miniseries Bose: Dead/Alive in 2017. She subsequently appeared in the film Nanu Ki Jaanu (2018) and the web series Main Hero Boll Raha Hoon (2021), Aar Ya Paar (2022), and IC 814: The Kandahar Hijack (2024). Patralekhaa is married to actor Rajkummar Rao.

Patralekha was born on February 20, 1990, in Shillong to a Bengali family. Her father was a chartered accountant, and her mother, Papri Paul, is a homemaker. In an interview, she revealed that her grandmother was a poet. She has two siblings, Parnalekha Paul and Agnish Paul. She attended The Assam Valley School (a boarding school) in Assam and then graduated from Bishop Cotton Girls' School in Bengaluru.

Patralekha's father wanted her to follow in his footsteps, but she was interested in acting. While graduating from the H.R. Directed by Hansal Mehta, the film tells the story of a poor couple from Rajasthan who come to Mumbai in search of employment. Made on a low budget, the film was a box office success. Sweta Kaushal of Hindustan Times wrote that Patralekhaa "played her character with utmost ease." For this film, she received the Screen Award for Most Promising Newcomer – Female.

Love Games (2016) - IMDb

Her next film was Love Games, an urban thriller directed by Vikram Bhatt and produced by Mukesh Bhatt and Mahesh Bhatt. Patralekhaa played a woman who is attracted to a man. However, the film received negative reviews and failed at the box office.

In 2017, Patralekhaa made her web debut with Bose: Dead/Alive, starring opposite Rajkummar Rao. The series earned her an iReel Award nomination for Best Actress – Drama . In 2018, she appeared in the series Cheers. She then played a ghost in Nanu Ki Jaanu, opposite Abhay Deol. However, many critics criticized her role in the film as "insignificant."

In 2019, Patralekhaa played a surrogate mother in the Zee5 film Badnaam Gali opposite Divyenndu Sharma and her performance received positive response.

After starring in the series Forbidden Love in 2020, in 2021, she appeared as an aspiring actress opposite Parth Samthaan in Main Hero Boll Raha Hoon. Archika Khurana of The Times of India wrote, "Patralekhaa oozes glamour as Laila and her chemistry with Parth is stellar." In 2022, she played an activist-doctor in the series Teerdaaz. Saibal Chatterjee of NDTV said, "Patralekhaa is impressive in the scenes that matter. I wish there had been more such moments; her performance would have gained more depth and range."
