Piyush Mishra was born on January 13, 1962, in Gwalior to Kumar Sharma. His childhood name was Priyakant Sharma. He was adopted by his father's elder sister, Taradevi Mishra, who had no children. His parents enrolled him in Carmel Convent School, Gwalior, so he could pursue his studies. However, Piyush was more interested in singing, painting, and acting than in his studies. He later attended J.C. Mills Higher Secondary School. Living in his aunt's strict environment fostered a rebellious nature, reflected in his first poem, "Zinda Ho Haan Tum, Koi Shak Nahin," which he wrote in eighth grade. In tenth grade, he changed his name to Piyush Mishra by submitting an affidavit to the Gwalior District Court.

It was during this period that he became interested in theatre. His talent was recognized on stages like Kala Mandir and Rangashree Little Ballet Troupe. Despite his family's insistence on focusing on his studies, he took the entrance exam for the National School of Drama (NSD) in 1983 and moved to Delhi. He graduated from NSD in 1986. While there, he composed music for his first play, "Mashrecki Hoor." In his second year at NSD, German director Fritz Bennewitz cast him in the lead role in Hamlet, which catapulted his acting career into a new light. In 1989, through his friend N.K. Sharma, he became acquainted with communist ideology and remained active in the leftist movement for nearly 20 years.

After graduating from NSD, Piyush Mishra began working as a theater actor in Delhi. In 1990, he founded the theater group "Act One" with artists like N.K. Sharma, Manoj Bajpayee, Gajraj Rao, and Ashish Vidyarthi. He wrote and directed several plays, most notably the famous play "Gagan Damama Bajiyo" (1994), based on Bhagat Singh.
In 1996, he joined the Asmita Theatre Group and performed his popular solo show, "An Evening with Piyush Mishra." He also received critical acclaim for his role as Suraj Singh in the play "Court Martial."
